1、紅茶提取物茶多酚產(chǎn)品規(guī)格: 30%,60% 茶多酚檢測(cè)方法: Folin D法有效成分: 茶多酚產(chǎn)品來(lái)源: 山茶科植物山茶的葉提取方法: 乙醇提取產(chǎn)品外觀: 棕黃色粉末1. 理化性質(zhì)分子結(jié)構(gòu)類(lèi)型:一類(lèi)多酚化合物溶解性:易溶于水及乙醇,味苦澀2. 質(zhì)量標(biāo)準(zhǔn)檢驗(yàn)2.1 鑒別:(對(duì)照品:沒(méi)食子酸)2.1.1 TLC檢查,與對(duì)照品Rf值、顏色一致2.1.2 IR檢查,與對(duì)照品指紋圖譜吻合(紅外吸收光譜)2.2 紅茶茶多酚的檢測(cè)方法:2.2.1 原理福林酚試劑氧化茶多酚中-OH基團(tuán)并顯藍(lán)色,最大吸收波長(zhǎng)為765nm, 用沒(méi)食子酸作校正標(biāo)準(zhǔn)定量茶多酚。2.2.2 試劑及設(shè)備1. 色譜純乙腈2. 甲醇1.
2、甲醇水溶液(體積比)=7:32. 碳酸鈉(Na2CO3)3. 福林酚試劑4. 10%福林酚試劑(現(xiàn)配):將20mL福林酚試劑轉(zhuǎn)移到200mL容量瓶中,用水定容并搖勻。5. 7.5% Na2CO3溶液(質(zhì)量濃度):稱(chēng)取37.50g0.01gNa2CO3(10.3),加適量水溶解,轉(zhuǎn)移至500mL容量瓶中,定容至刻度,搖勻。6. 沒(méi)食子酸標(biāo)準(zhǔn)儲(chǔ)備溶液(1000g/mL): 稱(chēng)取0.110g0.001g沒(méi)食子酸(GA,相對(duì)分子質(zhì)量188.14),于100mL容量瓶中溶解并定容至刻度,搖勻.7. 沒(méi)食子酸工作液:用移液管分別移取1.0,2.0,3.0,4.0,5.0mL的沒(méi)食子酸標(biāo)準(zhǔn)儲(chǔ)備溶液(10.8
3、)于100mL容量瓶中,分別用水定容至刻度,搖勻,濃度分別為10,20,30,40,50g/mL。2.2.3 樣品液配置準(zhǔn)確稱(chēng)取10g紅茶提取物樣品,用適量水溶解,轉(zhuǎn)入100mL容量瓶中,加水定容至刻度,搖勻,待測(cè)。 2.2.4 樣品含量的檢測(cè)用移液管分別取沒(méi)食子酸工作液、水及測(cè)試液各1.0mL于刻度試管內(nèi),在每個(gè)試管內(nèi)分別加入5.0mL的10%福林酚試劑,搖勻。反應(yīng)3min8min內(nèi),加入4.0mL7.5% Na2CO3溶液,加水定容至刻度、搖勻。室溫下放置60min。用10mm比色皿、在765nm波長(zhǎng)條件下用分光光度計(jì)測(cè)定吸光度(A)。根據(jù)沒(méi)食子酸工作液的吸光度(A)與各工作溶液的沒(méi)食子酸
4、濃度,制作標(biāo)準(zhǔn)曲線。 2.2.5 結(jié)果計(jì)算比較試樣和標(biāo)準(zhǔn)工作液的吸光度,按下式計(jì)算:式中:A樣品測(cè)試液吸光度;V樣品液體積;d稀釋因子(稀釋倍數(shù));SLOPEstd 沒(méi)食子酸標(biāo)準(zhǔn)曲線的斜率;m樣品干物質(zhì)含量,%;m1樣品質(zhì)量,單位為克(g)。2.3 檢查:干燥失重: 2%灰分: 0.5%重金屬: 百萬(wàn)分之五農(nóng)藥殘留: 百萬(wàn)分之二溶劑殘留:0.1%2.4 衛(wèi)生檢查細(xì)菌總數(shù):1000/g霉菌總數(shù):100/g大腸桿菌: 無(wú)沙門(mén)菌: 無(wú)2.5 粒度: 均勻接近細(xì)微顆粒2.6 包裝: 雙層朔料袋封口、外包裝紙桶、桶外印有品名、毛重、批號(hào)、生產(chǎn)單位、生產(chǎn)時(shí)間、有效時(shí)間、貯運(yùn)方法等。2.7 有效期: 2年2

Gallic acid working solution: accurately transfer 1.0, 2.0,3.0,4.0,5.0mL Gallic acid standard store solution into 100mL volumetric flask, and mix with water to volume. The concentration series are 10
12、, 20, 30, 40, 50g/mL.2.2.3 Preparation for test sampleAccurately taking 10g of Black Tea Extract samples dissolved in appropriate water, then transfer into 100mL volumetric flask, adding water to volume.2.2.4 Sample content testPartly taking 1.0mL Gallic acid working solution, Water, Test sample add
13、 into 3 graduated test tube. Each test tube be add into 5.0mL 10% Folin-Ciocalteu solvent, mixed. Let the liquid reaction 3min8min, then add 4.0mL 7.5% Na2CO3 solution and water to volume, mixed. Lay at room temperature. Detect the absorbance(A) by a 10mm cuvette at 765nm wave.Making the working cur
